马心肌血素中色氨酸残基的二维荧光相关光谱研究

【摘要】 光谱分辨率的提高与"序列顺序"法则被认为是广义二维相关光谱中的两大基本优势。然而,对广义二维相关光谱中的"序列顺序"法则却很少有人进行系统的研究。该工作在本研究小组理论研究的基础上,本文采用实验的方法-二维荧光相关光谱对马心肌血素(HM)中两种色氨酸残基(W7与W14)在不同浓度的NaI和丙烯酰胺(AA)猝灭剂存在下的强度变化规律进行分析,从而对广义二维相关光谱中的"序列顺序"法则所提供信息的准确性与可靠性进行实验验证。结果发现,与我们的理论研究结果一样,运用"序列顺序"法则在某些情况下并不能提供准确可靠的变化顺序或变化速率的信息。
【Abstract】 Band resolution enhancement and the"sequential order"rules are the two primary advantages of generalized two-di- mensional(2D)correlation spectroscopy.No systematic study on the latter was found,however.Based on our previous theoreti- cal and simulation studies,an experimental study was carried out to test the validity of the"sequential order"rules,using 2D fluorescence correlation spectroscopy to analyze the two tryptophan residues in horse heart myoglobin(HM)under different con- centrations of NaI or acrylamide quencher.It was concluded,as also found from our theoretical analysis,that the"sequential or- der"rules do not always provide correct information on the local sequential order or rate difference of two events in generalized situations.
