桑色素合钴(Ⅱ)配合体系与DNA作用的热力学稳定性研究

【摘要】 用分光光度法研究了桑色素-Co(Ⅱ)配合体系与DNA作用的热力学稳定性。Co(Ⅱ)使桑色素380nm(pH=5.3)处的吸收峰红移到400nm附近,且吸光度增大。DNA的加入可使桑色素-Co(Ⅱ)在400nm的吸收峰移动至421nm附近使桑色素-Co(Ⅱ)配合物的吸光度增强。测定结果显示,在25-50℃范围内,Co(Ⅱ)与桑色素可以生成稳定的配合物。反应体系简单,反应条件容易实现,适于大规模合成应用。DNA的加入量与配合物特征峰吸光度的增加值在一定范围内呈正比关系。表明桑色素-Co(Ⅱ)配合物与DNA存在相互作用,可作为探针分子定量测定实际样品中DNA,基于此,建立了在较宽范围内用分光光度法测定DNA的方法和研究配合物热力学稳定性的方法。

【Abstract】 This paper reports the interaction of Co(Ⅱ) complex with DNA by spectrophotometry. Composition ,stability constant are determined by the absorbance. Addition of Co(Ⅱ)make the absorption peak of Morin at 380nm (pH=5.3) shifted to 400nm , addition of nucleic acids to the Co(Ⅱ)―Morin complex leads a decrease of the absorption at 380nm and an increase at 421nm .The increase of absorbance of the complex are proportional to the amount of nucleic acids added in a certain concentration range. Experimental results indicate that Co(Ⅱ) complex has interacted with DNA. Based on this, a new sensitive nucleic acids analysis method is established. The complex of Co(Ⅱ)-Morin can be used as a probe to determine the amount of DNA in a wide rang by spectrophotometry.
