蛋白质与小分子相互作用的动力学毛细管电泳研究

【摘要】 <正>采用动力学毛细管电泳(kinetic capillary elec- trophoresis,KCE)研究亲和相互作用是一种新方法。它依据电泳淌度差异的拆分作用与分子间的作用平衡。所以采用动力学毛细管电泳中可以获得的参数包括了 kon、koff和 kd、△S、△H等。对于—个分子相互作用体系,靶体和配体之间为特殊的强的非共价结合,并且其中的一个为生物高聚物或大分子,比如:蛋白质分子、DNA 和 RNA 等。
【Abstract】 Kinetic capillary electrophoresis (KCE) is a new method for interaction studying.It depends on the split function of different electrophoresis velocities and the equilibrium of the interaction.So many parameters including kon,koff,and kd,△S,△H all can be obtained by KCE.KCE is a conceptual platform for kinetic homogeneous affinity methods,and furthermore it is a conceptual studying platform.But the applications and method of KCE need to be studied more deeply.Our group has developed reported method,technology and calculation.In our method,the concentration of substance cannot be tagged and so on.Three kinds of kinetic capillary electrophoresis have been used in interaction analysis of serum albumin with drugs and other aromatic environmental polluting compounds in our group. The results showed that more affinity information could be obtained and it might be developed further more.We have also compared these methods of kinetic capillary electrophoresis with other interaction analysis methods such as fluorescence,conventional capillary electrophoresis,electrochemical methods.These methods were depended on the temperature and buffer constitutes,but they were free of the inner diameter and length of capillary.The kinetic capillary electrophoresis could be used in screening and evaluation of activity of drug molecules and toxicity of pollutants.
