几种富勒烯衍生物光致细胞膜毒性的动态研究

【摘要】 利用荧光显微镜研究了在显微镜汞灯绿光激发下二加成亚甲基富勒烯 [60]二膦酸四乙酯(BFDP)、二加成富勒烯[60]丙二酸(DMAC60)和富勒醇(Ful- lerol)3种不同结构富勒烯衍生物对 HeLa 细胞细胞膜的光诱导损伤作用,发现 BFDP、DMAC60对 HeLa 细胞细胞膜的光诱导损伤过程在低浓度下(4μm/L)即可发生,且有浓度依赖性,而富勒醇(Fullerol)在相同光照条件下并不引起细胞膜损伤。对机理初步研究发现 BFDP、DMAC60光诱导膜损伤过程有活性氧的参与,但涉及活性氧的种类不同。这些结果直接证实细胞膜是 C60衍生物的光敏作用靶点,并提示活性氧可能参与这一过程。
【Abstract】 By using fluorescence microscope,the photo-induced damage to cell membrane of HeLa cells by bisadduct of tetraethyl methano [60] fullerenediphosphonate (BFDP),dimalonic acid C60 (DMAC60) and Fullerol was investigated in the presence of green light of mercury lamp.The data showed that the photo-induced toxicity against cell membrane activity occuned even at the presence of low concentration of (4 μm/L),and the damage was dose-dependent, but the Fulllerol can not induce any damage to cell membrane of HeLa cells at the same conditions.The results demonstrated that photo-induced cell membrane damage of BFDP and DMAC60were related to the produce of reactive oxygen species,and involved different species.It is concluded that cell membrane was a target of photosensitive action produced by BFDP and DMAC60,and implied that reactive oxygen species was probably involved in the photosensitive action.
