水溶性富勒烯[60]多羟基衍生物富勒醇光诱导DNA剪切及其细胞毒性的研究(英文)

【摘要】 利用质粒 DNA(pXJ41-neo)及 HeLa 细胞做实验对象,研究了水溶性富勒烯[60]多羟基衍生物富勒醇光诱导 DNA 剪切及其细胞毒性作用。采用琼脂糖凝胶电泳和 MTT 法分别检测受作用后的 DNA 和 HeLa 细胞。结果表明,富勒醇能剪切 DNA 并抑制细胞生长,且具有光辐射及浓度依赖性。在500 W 碘钨灯距离20 cm 处光照1.5 h 的条件下,0.4 mM 富勒醇几乎能把所有的 DNA 从原始超螺旋形式剪切成线性形式,同样在光照条件下,20 μM富勒醇能显著抑制 HeLa 细胞生长。一些活性氧清除剂包括叠氮化钠、甘露醇和超氧化物歧化酶均可抑制光诱导下富勒醇对 DNA 的切割作用。这一结果进一步证实了富勒醇的光诱导生物活性,而且为富勒烯衍生物应用于肿瘤的光动力治疗提供了新的途径。

【Abstract】 This report aims to study their bioactivities of DNA cleavage and cytotoxicity of polyhydroxylated [C60]fullerene derivatives fullerols,using a plasmid pXJ41-neo DNA and HeLa cells as experimental materials.Cleaved DNA products were detected by agarose gel electrophoresis,and cell growth was determined by MTT reagent-based colorimetric assay.The results showed that fullemls could stimulate DNA cleavage and inhibit cell growth,and both activities were dose and irradiation dependent.0.4 mM Fullerols together with 1.5 h exposure to a 500 W tungsten halogen lamp with a distance of 20 cm could convert most of plasmid DNA from the intact form into cleaved forms.In the presence of irradiation,cell growth was significantly inhibited by 20 μM fullerols.Scavengers of various reactive oxygen species (ROS) including azide,mannitol and superoxide dismutase (SOD) could inhibit the photoinduced DNA cleavage of fullerols.These data provided some novel evidences on the photoinduced biological activities of fullerols,and implied a possible use of these fullerene derivatives in the photodynamic therapy (PDT) of tumors.
