转铁蛋白受体介导的GaC-Tf和Ga2-Tf内吞与释放

【Abstract】 There has been an increasing interest in the use of gallium in anticancer activity. However, whether the uptakes of two species of transferrin including digallium transferrin (Ga2-Tf) and the C-terminal monogallium transferrin (GaC-Tf) by cells are differenrt is not well understood. In this work the mechanism of both passing in and out K562 cells has been established by using 125I-labeled transferrin. At 0 ℃ the association constants for the binding of both to K562 cells were found to be similar. There were about (1.5±0.08)×105 binding sites per cell surface. Pretreated cells with pronase abolished the binding of both significantly. The transferrin species did not affect the rate for internalization and release in the cell type at 37 ℃. The results indicated that ransferrin receptor plays an important role for the endocytosis and release of GaC-Tf and Ga2-Tf and the both processes are regardless of the gallium content of the protein by K562 cells.
