6-二甲氨基嘌呤诱导三倍体合浦珠母贝的研究(Ⅰ)

【摘要】 一种新的诱导剂6-二甲氨基嘌呤(6-DMAP)抑制第二极体诱导合浦珠母贝 Pinctada martensii(D.)三倍体。对6-DMAP 浓度,处理持续时间及起始时间进行了研究。结果表明孵化率与这三个参数呈负相关,而三倍体诱导率在一定范围内呈正相关。合适的6-DMAP 浓度为60~80mg.L-1,处理持续时间约20min,起始时间的选择受水温及卵发育程度的影响。75mg.L-1 6-DMAP 与0.5mg.L-1细胞松驰素 B (CB)处理对比,6-DMAP 处理组的孵化率(4.57%)明显高于 CB 处理组(2.23%),而三倍体诱导率(90.00%)与 CB 处理组(93.55%)差别不明显。由于6-DMAP 具有低毒、高效、易于操作的特点,是一种优良的多倍体诱导剂。
【Abstract】 In this study,6-dimethylaminopurine(6-DMAP),a new inductor,was used to inhibit the sec- ond polar body in order to induce triploid pinctada martensii (D.).The effects of 6-DMAP concen- tration,treatment duration and timing of treatment after fertilization on the normal D-shaped larvae ratio and the percentage of triploids were examined.The results indicated that the survival to D- stage was inversely related to 6-DMAP concentration,treatment duration and time of treatment after fertilization;and that the percentage of triploids was related to these three parmeters.Optimal 6-DMAP concentration was 60~80 mg.L-1.Optimal treatment duration was about 20 min.The timing of treatment application was dependent on the temperature the of seawater and the develop- mental condition of the Oocytes.Treatment with 75mg.L-16-DMAP resulted in 4.57% survival rate to D-stage.The rate of survival to D-stage was greater than 2.23% for the group treated with 0. 5mg.L-1CB.There was no significant difference in the percentage of triploids induced by treatment with 6-DMAP (90.00%) with 6-DMAP(90.00%),or with CB (93.55%).6-DMAP treatment is a new method that is easy to handle,and is a less dangerous,most efficient technique to induce triploid Pinctada martensii (D.).
