4个Y STRs基因座的复合扩增

【摘要】 目的建立DYS19、DYS389 Ⅰ、DYS389Ⅱ、DYS385复合扩增体系。方法遴选Y STRs基因座的引物,分别用FAM、TAMRA、TET标记DYS19、DYS385、DYS389 Ⅰ、DYS389Ⅱ,优化扩增条件,考察扩增体系的个体识别能力、灵敏度、种属特异性及突变情况。结果所建立的4基因座Y STRs复合扩增体系分型清晰,单倍型多样性达0.989,且特异性好,灵敏度高(lng DNA),未观察到突变。结论所建立的4个Y STRs基因座复合扩增方法适合法医学应用。
【Abstract】 Objective A multiplex polymerase chain reaction (PCR) assay capable of simultaneously amplifying multi-Y chromosome short tandem repeat (STR) markers was developed to aid forensic identity testing. Methods Efforts were made to establish Y chromosome STR tetraplexes which include the loci DYS19, DYS389 Ⅰ, DYS389 Ⅱ and DYS385 through careful design of primers, examination of known allele ranges and optimization of PCR condition. Series experiments were carried out to test the specificity and sensitivity of the tetraplexes. Results The tetraplexes can exactly type Y STRs and be of good specificity and sensitivity. 100 genotypes of 4 Y STRs loci were found in 111 Guangzhou Han people and haplotype diversity(HD)=0.989. Conclusion The tetraplexes of Y STRs can be used in forensic identification cases.
