氧气对316L不锈钢表面等离子体聚合甲基丙烯酸丁酯涂层性能及内皮细胞生长行为的影响

【摘要】 为了加速316L不锈钢支架表面的内皮化,本文首次采用等离子体聚合沉积技术在其表面制备聚甲基丙烯酸丁酯涂层。重点研究了氧气对制备涂层性能(化学组成与结构、形貌和亲水性)以及内皮细胞生长行为(细胞粘附、细胞生长等)的影响。反射红外光谱(ATR-FTIR)和X-光电子能谱(XPS)分析结果显示,氧气并没有明显改变聚甲基丙烯酸丁酯的化学组成和结构,涂层中含氧基团的数目并未显著提高。SEM和接触角测试结果表明,氧气的引入提高了涂层的光滑性和亲水性。MTT和SEM实验表明,聚甲基丙烯酸丁酯涂层,尤其是在有氧存在条件下,可有效促进内皮细胞的粘附、生长,是制备具有原位诱导内皮组织再生功能支架的一种有效手段。

【Abstract】 To promote the re-endothelialization of stent surface, poly n- butyl methacrylate (PPBMA) thin films were deposited by radio-frequency plasma technique for the first time. The effects of oxygen on the chemical structure, morphology, hydrophilicity of PPBMAs and attachment, proliferation of endothelia cells on the PPBMAs coating were carried out. ATR-FTIR and XPS showed that oxygen had little influence on the chemical structure and composition of the PPBMAs. The resultant PPBMA films deposited without and with oxygen all had relatively strong absorption from C=C bonds and-OH bands, but almost no absorption of C=O and C-O bands. SEM and static contact angle measurement indicated that in case of deposition with oxygen, the smoothness and hydrophilicity of the PPBMA were dramatically improved. MTT and SEM revealed that the PPBMAs coating, especially deposition in the case of oxygen, could effectively promote the attachment and proliferation of the endothelia cell.
