新型磷灰石/硅灰石生物活性玻璃陶瓷多孔支架材料的细胞相容性研究

【摘要】 种子细胞、支架材料和二者的相互作用是骨组织工程需要解决的三大问题。在材料方面,研制力学性能和生物相容性均好的可降解多孔支架材料一直是研究者们努力的方向。本研究采用溶胶凝胶法制备前驱体粉末,以硬脂酸为成孔剂,经混料、模压成型、干燥及高温烧结步骤制备AW生物活性玻璃多孔支架材料,其主主晶相为磷灰石和硅灰石。采用体外模拟体液(SBF)实验评价支架的生物活性和生物降解性。采用经成骨诱导的大鼠骨髓间充质干细胞(rMSC)为种子细胞,与AW支架材料进行体外复合培养,通过扫瞄电镜观察和MTT比色法分析细胞在材料上的粘附、增殖情况。结果表明AW生物活性玻璃在体外模拟体液中第7天其基体表层局部生成大量细粒状的羟基磷灰石晶粒;AW支架材料具有良好的生物相容性,rMSC能在支架材料上粘附增殖。提示该种新型支架材料有望成为具有发展潜力的骨组织工程支架材料。

【Abstract】 There are three key factors in tissue engineering: seeding cells, scaffold and their interaction. For materials, the degradable porous scaffolds which have good mechanics and good biocompatibility are preferred. AW-GC was prepared by sintering the precursor powder derived from sol-gel process. Porous AW scaffold, including apatite and wollastonite, was prepared through mixing, using stearic acid as porogen, compression molding and sintering process. The bioactivity and biodegradation were investigated by soaking scaffolds in simulated body fluid (SBF). Osteo-induced rat mesenchymal stem cells (rMSC) were co-cultured with scaffold in vitro. The cell-material complex was observed by SEM, the assay of MTT to evaluate the adhesion and proliferation of the cells. The results showed that a layer of carbonate hydroxyapatite (HCA) was formed on the surface of scaffolds when soaked in SBF, which indicated they were bioactive; Osteo-induced rMSC was biocompatible with scaffold, and could attach, proliferate on scaffold, which indicated the scaffolds had potential for application in bone tissue engineering.
