聚乳酸/丝素复合材料的制备和肝细胞的生长性能

【摘要】 将不溶性的丝素微球加入聚乳酸溶液,使用冻干法制得聚乳酸/丝素复合支架材料。电镜结果显示复合材料形成了良好的三维支架结构,丝素微球均匀地分布在聚乳酸支架基体内。细胞结果表明HepG2细胞在三维材料表面黏附和伸展状况良好。同聚乳酸支架相比,细胞在复合支架上的生长情况有了显著提高,细胞能相互作用,形成大于100微米的聚合体。考虑到聚乳酸能形成良好的三维支架结构,而丝素能提高细胞的相容性,聚乳酸/丝素复合材料将成为一种非常有前途的组织支架基体。

【Abstract】 A composite scaffold composed of polylactic acid (PLA) and fibroin was prepared by adding insoluble fibroin microspheres into PLA solution. Observation of the PLA/fibroin composite scaffold by scanning electron microscopy (SEM) showed that fibroin microspheres uniformly distributed in PLA matrix and interconnected pore structures were formed in the scaffolds. Cell test showed that the HepG2 cells attached and spread well on the surfaces of the composite scaffold. Compared with that on PLA scaffolds, the attachment and growth of hepG2 on the composite scaffold were greatly improved. HepG2 cells attached firmly with the hybrid sponges and established cell-cell contacts to form aggregates with a size up to 100μm. Considering that the PLA skeleton facilitated the desired shape formation, and fibroin microspheres improved the cell growth, the PLA/fibroin composite scaffold would become the promising matrix for hepatic tissue engineering.
