日本对虾肌肉中SOD分离条件的探索

【摘要】 为了更好的从日本对虾肌肉中提纯超氧化物歧化酶(SOD),本实验对SOD纯化过程中所用匀浆缓冲液的pH值、热处理温度、盐析和柱层析等分离条件进行了摸索,并对其进行了初步的分离纯化。发现匀浆缓冲液的pH值为7.8,热处理温度为65℃,硫酸铵饱和度为70%-90%时提纯条件最佳。结果获得较纯的SOD,比活力为488.04,纯化倍数为39.30

【Abstract】 Superoxide dismutase (SOD) is known to be major scavengers of superoxide radicals produced in the body of organisms. In this paper, the purifying method of SOD from muscle tissue of the shrimp Penaeus japonicus was studied. When the pH of extract buffer was at 6.5, the highest activity of SOD was achieved. The heat treatment of 65 ℃ and 70% - 90% saturated ammonium sulfate could fractionated the protein. The active fraction yielded by DEAE-32 column chromatography under eluting buffer, pH 7.8 was collected, SOD at higher purity was primarily obtained.
