几种不同进化程度动物细胞蛋白二硫键异构酶活性及含量研究
THE CONTENT AND ACTIVITY OF PDI IN DIFFERENT EVOLUTION ANIMAL CELLS

【摘要】 使用Hillson方法及酶联免疫技术,测定了几种不同进化程度动物细胞蛋白二硫键异构酶的活性及其含量.结果表明,在不同进化程度的动物细胞内均有其分布,但酶活性及含量差异显著.在高等脊椎动物肝细胞中,其含量约占细胞总蛋白的2%,酶活性约为4U/g,与低等脊椎动物和无脊椎动物肝细胞差异显著.我们还系统比较了不同进化程度动物肝细胞的微粒体获得率及蛋白含量,得到了不同细胞微粒体含量明显不同,单位重量微粒体蛋白含量基本相同的结果,表明不同细胞内质网含量不同,而单位内质网蛋白种类和含量具有一定保守性.
【Abstract】 Protein disulfide isomerase (PDI)is one of main retention proteins in ER. Its main biological function is catalysis for disulfide formation of nascent membrane and secretory proteins and it also plays a role of molecular chaperone. By the method of Hillson’s and ELISA, the content and activity of PDI from different evolutionary animal cells were first determined. The result is that PDI distributes in every evolutionary animal cells, though the activity and content may be very different. The weight and protein content of the microsome from different evolutionary animal are also assayed, and the weight of microsome varies obviously and the protein content of microsome in unit is almost the same.
