金鱼四个代表品种基因组DNA的遗传变异研究

【摘要】 对四个金鱼代表品种进行了随机扩增多态DNA分析.用45个10bP的随机引物对实验鱼的基因组DNA进行扩增,平均每个品种金鱼获得225个标记,每个引物平均提供5.0个标记.RAPD标记共享度计算结果显示:金鱼各品种间共享度指数最大值为86.78%,最小值为82.70%,平均值为84.53%;金鱼的遗传变异指数为0.2312.这些结果表明:金鱼的四个代表品种的基因组DNA的相似性高,遗传变异程度低.

【Abstract】 Random amplified polymorphic DNA (RAPD) technique were used to investigate genetic variation of four representative varieties of goldfish. Using 45 single 10 base-long primers of arbitrary DNA sequence, on average, about 225 RAPD markers were observed in each variety. The markers which provided by a single primer were 5. The average, maximum and minimum proportion of RAPD bands shared among four varieties of goldfish were 84. 53%, 86.78% and 82.70%, respectively. The index of genetic variation was 0.2312. These results indicated that the similarities of genomic DNA were high and the degree of genetic variation was low in the four representative varieties of goldfish.
