赣江水体有机提取物的雌激素活性及遗传毒性研究

【摘要】 目的探讨赣江水体有机提取物的雌激素活性及遗传毒性,从而为赣江水体污染的环境风险评价提供科学的理论依据。方法分别从赣江的上游、中游和下游各3个断面的水体采集水样,接着采用XAD-2大孔树脂对水样中的有机物进行浓缩提取,然后利用子宫增重实验检测有机提取物的雌激素活性,并利用微核实验检测有机提取物的遗传毒性。结果各水样有机提取物均显示出雌激素活性和遗传毒性,且具有剂量-反应关系,与溶剂对照组比较,差异具有统计学意义(P<0.01);不同采样断面水样有机提取物的雌激素活性有所不同,总体趋势是下游>中游>上游;下游水样有机提取物所致微核率明显地高于上游和中游水样(P<0.01);而上游水样有机提取物所致的微核率与中游之间的差异则无统计学意义(P>0.05)。结论赣江水体己受到环境内分泌干扰物的污染并具有一定的遗传毒性,对环境及人群健康具有潜在的风险。

【Abstract】 Objective To investigate the estrogenic activity and genotoxicity of organic extracts from water of Ganjiang River,and provide theoretically scientific basis for further environment risk assessment of Ganjiang River water pollution.Methods Water were sampled with amberlite XAD-120 separately at three site that located at upper,middle and down section of Ganjiang River,then the estrogenic activity of organic extracts were determined by uterotrophic assay in vivo;and the genotoxicity of such compounds were detected by micronucleus test. Results It was show that the estrogenic activity and genotoxicity of organic extracts from water of Ganjiang River were found in all samples,which were significantly higher than those of the vehicle controls (P<0.01),and there was positive dose-response relationship.Organic extracts at different section have showed different estrogenic activity,the order of estrogenic activity of those samples is as follows:upper section>middle section>down section.Micronucleus rate in the groups treated with organic extract of down section water sample were higher than those of middle and upper sections with significant difference(P<0.01);however,there was no difference between upper and middle sections. Conclusion It can been concluded that waters in Ganjiang River have been contaminated by endocrine disrupting chemical and have showed mutagenic effect in some extent,which might pose potential hazards to environment and population health.
