微囊藻毒素抑制小鼠淋巴细胞免疫功能的研究

【摘要】 目的探讨微囊藻毒素LR(ME—LR)抑制小鼠脾淋巴细胞免疫功能的可能机制。方法分离小鼠脾淋巴细胞,分别给予0、1、3、5、10μg/ml的MC-LR体外染毒,培养24h,收集上清液,ELISA法检测淋巴细胞因子IL-2的产量,同时收集淋巴细胞,流式细胞仪测定淋巴细胞凋亡率;培养72h。MTT法检测MC-LR对小鼠脾淋巴细胞增殖的影响:。结果各剂量组的MC-LR均可使小鼠脾淋巴细胞增殖指数降低(P<0.05);染毒剂量≦3μg/ml的MC-LR可刺激脾淋巴细胞分泌IL-2,而高于5μg MC-LR/ml组却降低脾淋巴细胞IL-2的分泌量,呈双相性变化;10μg/ml的MC-LR迅速升高脾淋巴细胞凋亡率(P<0.05)。结论体外培养条件下,微囊藻毒素MC-LR能明显抑制小鼠脾淋巴细胞增殖,降低淋巴细胞因子IL-2的产量并诱导淋巴细胞凋亡,其中抑制脾淋巴细胞增殖作用尤其明显。
【Abstract】 Objective Study the possible mechanism on the lymphocyte immunologic function of mice inhibited by microcystin-LR.Method Mice splenic lymphocytes were cultured with MC-LR at the doses of 0,1,3,5,10μg MC-LR /ml in vitro,the production of lymphocyte IL-2 was test by ELISA assay and apoptosis of splenic lymphocytes were monitored by flow cytometry after incubation with MC-LR for 24h.The lymphocyte proliferation was determined after the cells treated by MC-LR for 72h.Results In comparison with controls,the lymphocyte proliferation was significantly lower in MC-LR treated groups (P<0.05).The production of lymphocyte IL-2 was stimulated by MC-LR in doses≦3μg/ml but was inhibited in doses≧5μg/ml,and the apoptosis of splenic lymphocytes were significantly increased in dose of 10μg/ml (P<0.05).Conclusion MC-LR could decrease the production of mouse lymphocyte IL-2 and increase the apoptosis of the lymphocytes in vitro.MC-LR could inhibit lymphocety proliferation of mice especially.
