“川草2号”老芒麦愈伤组织再生系统的研究(英文)

【摘要】 以“川草2号”老芒麦的成熟种子为外植体,MS并添加2,4-D5.0 mg.L-1和Kinetin0.05 mg.L-1为基本培养基,26℃暗培养40 d左右,可诱导愈伤组织产生。愈伤组织继代培养8周,形成易于再生的致密、黄白色的愈伤组织。实验表明, 成熟胚诱导的愈伤组织有较高的分化率,达到54%。诱导愈伤组织的最适培养基为MS+2,4-D5.0+Kinetin0.05,诱导率达到85%以上;分化培养基为1/2MS+Kinetin0.1+2,4-D1.5+20 g.L-1蔗糖,分化率在54%以上;生根培养基为不含任何生长调节剂的1/2MS,生根率达到97%。高效而稳定的“川草2号”老芒麦愈伤组织再生体系的建立,为研究“川草2 号”老芒麦的新品种选育和遗传转化打下基础。
【Abstract】 In order to realize the quick propagation of Elymus sibiricus L.cv. ’chuancao No. 2 ’ and make it to be a sensitive material for its gene transformation,we have made a set of effective and stable callus regeneration system of Elymus sibiricus L.cv. ’chuancao No. 2’ . The mature seeds of Elymus sibiricus L.cv.’chuancao No. 2’were cultured on Murashige and Skoog(MS) medium supplemented with 5.0 mg·L-1 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.05 mg·L-1 kinetin in the dark at 26℃, the calluses were produced. Plants regenerated from whitish-yellow-coloured compact nodular callus formed after subculturing for 8 weeks. Higher frequency (54%) of shoot differentiation was obtained from the embryo tissues of mature seed when these calluses were cultured on plant regeneration medium containing half strength MS salts supplemented with 0.1 mg·L-1 kinetin,1.5 mg·L-1 2,4-D and 20g·L-1 sucrose. The green plants were rooted within six weeks in the root regeneration medium, and over 97% of these soil-established plants were obtained in the greenhouse when potted in a sand and peat mixture medium.
