昆虫病原线虫共生菌的杀虫活性

Insecticidal Activity of Symbiotic Bacteria from Entomopathogenic Nematodes

【摘要】 自9个昆虫病原线虫品系离体培养得到99株共生细菌菌株,通过生物测定从每个共生菌品系中筛选出高毒力菌株,比较后得到高毒力共生菌品系Ssy3及高毒力菌株Ssy3-9B。此菌株对玉米螟、小菜蛾、甜菜夜蛾、粘虫、黄粉虫幼虫有较高的胃毒毒力,对未死虫的生长发育有明显的抑制作用,对大蜡螟也具很高的血腔毒力。对高毒力菌株相关因子的研究表明:高毒力菌株及其杀虫粗提物经50℃处理10min及18W紫外灯下60cm高度垂直照射60min,杀虫活性仍很稳定:Ⅰ、Ⅱ型共生菌的胃毒毒力和血腔毒力无明显差别。

【Abstract】 Xenorhabdus and Photorhabdus are entomopathogenic bacteria symbiotically associated with insect pathogenic nematodes. They are highly virulent to insects. 99 strains of symbiotic bacteria were isolated from 9 strains of entomopathogenic nematodes. Bioassays showed the insec-ticidal activities varying according to species and isolates. The highly virulent strains were selected through a large number of bioassays. Xenorhabdus sp. sy3 has oral insecticidal activity. This bacteria was highly virulent to Galleria mellonella, Plutella xylostella, Tenebrio molitor, Latphygma exigua , Pseudaletia separata and Ostrinia furnacalis larvae. The insecticidal toxin was extracted primarily. The insecticidal toxin inhibited the growth of Ostrinia furnacalis . This bacteria and its insecticidal toxins were substantially heat stable to 50℃, and also stable to 60min ultraviolet ray. Ssy3 is a potential biocontrol agent. Phase variation is a feature to entomopathogenic nematodes bacterium. The insecticidal activities of phase Ⅰ and Ⅱ of Ssy3 were studied, and showed both had highly oral and haemocoel insecticidal activity.
