家蚕胚胎血淋巴保幼激素结合蛋白肽质量指纹图谱分析

【摘要】 采用基质辅助激光解吸电离飞行时间质谱对催青期出现的表达量较大的519号特异蛋白斑点进行了肽质量指纹图谱分析,得到了该蛋白斑点的肽质量指纹图谱。获得的肽质量指纹图谱与蛋白质数据库中已有的蛋白质酶解理论肽质量数或序列相比较,初步鉴定为血淋巴保幼激素结合蛋白。结合胚胎发育的进程,对该特异蛋白斑点进行了功能推测。血淋巴保幼激素结合蛋白在戊(?)以后的胚胎中存在且作用时间较长,控制着胚胎发育的进程,可以认为它是催青期胚胎发育过程中的—个极其重要的蛋白质。通过对特异蛋白斑点的质谱鉴定,从蛋白质的变化可以了解家蚕催青期结构基因激活表达变化的信息,进行基因功能推测。
【Abstract】 Using Matrix assisted laser desorption ionization time of flight (MALDI-TOF-MS) to analyzepeptide mass fingerprinting (PMF) of 519 special protein spot which were highly expressed during incubationstages,peptide mass fingerprinting of this protein spot was obtained.By comparing the peptide massfingerprinting with theoretic peptide quantity of protein enzymolysis or sequence existing in protein database,theprotein spot had been primarily identified as hemolymph juvenile hormone binding protein (hJHBP).Based onembryo development,the main functions of the protein spot was guessed in detail,hJHBP existed in the embryoafter late bulge stage,kept effective for a long period,and controlled the process of embryo development,therefore it could be considered as vital protein in the process of embryo development.Through Matrix assistedlaser desorption ionization time of flight (MALDI-TOF-MS)mass identification of special protein spots,information of structural gene activation and expression could be understood and the function of these genes inincubation stage could be speculated on the changes of proteins.
