茶树对茶尺蠖取食诱导的基因表达谱差异初探

【摘要】 基因表达谱差异显示技术是最近兴起的研究同种细胞在不同状态下基因表达差异的一种有效手段。目前已成功应用于植物对害虫取食诱导防御的分子机制和发掘植物内源抗虫基因的研究领域,并显示出独特的优势。本文利用DDRT-PCR技术,初步研究了茶树被害虫茶尺蠖取食后基因表达谱差异。结果表明:接头引物A3、A4、A7 和A8分别与随机引物R1-R8,R12组合扩增后,总共得到122条差异片段,占总条带数的22.6%。因此茶树被茶尺蠖取食后,能够诱导基因表达谱的差异。

【Abstract】 Techniques of differential display for gene expression profile, recently developed, are effective and practicable to explore gene expression and used in the fields of plant- insect interactions and searching insect-resistant genes successfully. DDRT-PCR was used to explore the differential gene expression profiles analysis of tea plant induced by Ectropic obliqua attack. The results were showed that 122 differential bands were obtained, which was 22.6% of total bands, by amplified in anchor primers A3, A4, A7, A8 and random primers R1-R8, R12, respectively. This suggested that the differential gene expression profiles were induced by Ectropic oblique in tea plant.
