乙醇对大鼠枯否细胞分泌TNF-α和微管表达的影响

【摘要】 目的:观察乙醇体外对培养枯否细胞分泌TNF-α和微管表达的影响。方法:体外培养枯否细胞中加入不同剂量(200 mg%、400 mg%、800 mg%)乙醇3 h或(和)24 h后,分别测定枯否细胞分泌TNF-α的能力和细胞微丝表达的强度.结果:乙醇作用3 h,200 mg%组培养上清中TNF-α含量显著高于正常对照组,400 mg%组和800 mg%组均显著于200 mg%剂量组:乙醇作用24 h,200 mg%和400 mg%组培养上清中TNF-α含量显著低于常对照组和同剂量3 h乙醇组;乙醇作用3 h,200 mg%组枯否细胞微管荧光灰度显著高于正常对照组和400 mg%组。结论:小剂量乙醇可以增强体外枯否细胞分泌TNF-α的能力,乙醇大剂量或长时间作用反而抑制枯否细胞分泌TNF-α的能力;小剂量乙醇可以增强微管表达,但大剂量乙醇则使其微管表达减速弱。
【Abstract】 To investigate the effect of ethanol on TNF-αsecrection and tublin expression of Kupffer cell in vitro.After kupffer cell’s having been cultured with ethanol added DMEM medium (200mg%, 400mg% or 800mg%) for 3 hours and (o024 hours,the TNF-αsecretion of Kupffer cells was tested and the tublin density of monoclonalanti-αTubulin-FlTC stained kupffer cells was determinated.The TNF-αsecretion of Kupffer cells in 200mg% ethanol added and 3 hour cultured group was significantly higher than in control,800mg% ethanol added groups.The TNF-αsecretion of Kupffer cells in 200mg% and 400 mg% ethanol added and 24 hour cultured groups was significantly lower than in 3 hour cultured groups.The tublin density in 200rag% ethanol added and 3 hour cultured group group was significantly higher than control and 400 rag% ethanol added groups.Ethanol in the lower dose can enhances TNF-αsecretion of Kupffer cells in vitro,but in a higher dose or for a long time culture,it can inhabit the TNF-αsecretion of Kupffer cells.Ethanol in the lower dose can increase tublin expression of Kupffer cells in vitro,but in a higher dose it can decrease the tublin expression of Kupffer cells.
