【摘要】 病理石蜡切片检查为禽骨髓细胞瘤病(ML)的肉种鸡病料(肝),经电镜检查可见疑似禽白血病病毒(ALV)粒子.为进一步确定此病毒确实为ALV-J病毒,我们以ALV-J原型株HPRS-103囊膜gp85基因为基础设计引物,体外扩增(PCR)gp85基因,获得3株病毒,分别为NM0138、NM0139和NM0140.将NM0138 PCR产物进行分子克隆和序列测定,结果表明,NM0138 gp85蛋白的推测氨基酸与ALV-J原型株HPRS-103、山东SD9901株、美国ADOL-R5-4株的gp85蛋白及内源性病毒EAV-HP env蛋白的氨基酸同源性分别为97.40%、95.13%、86.69%和85.06%.其中与HPRS-103的同源性最高,与EAV-HP同源性最低.因此可以确定本研究所克隆的基因确实为ALV-J病毒gp85基因,故暂命名为ALV-J-NM138.这为进一步研究本病打下了坚实的基础,同时也确定了PCR方法诊断禽骨髓细胞瘤病的特异性和实用性.更多还原

【Abstract】 After diagnosing the meat - type breeder chicken illness as avian myelocytomatosis by pathology, we discovered a kind of uncertain ALV virus in liver under electronic microscope (another article) . In order to attest this virus was ALV - J. we designed a pair of primer on the bese of HPRS - 103 gp85 gene and succeed amplified this gene in vitro. This three strains virus was named NM0138, NM0139 and NM0140 respectively. We cloned and sequencedNM0138 strain gp85 gene’ PCR product, and compared the predicted amino acid with those of HPRS - 103, SD9901, ADOL - R5 - 4 and EAV -HP. The results indicated that the predicted aminoacid sequences of NM0138, SD9901, ADOL- R5 -4 and EAV - HP gp85 protein had 97.4%,95.13%, 86.69% and 85.06% homology,respectively. Among them, the highest homology is HPRS - 103, and the lowest homology is EAV - HP. It is the reason that the gene that amplified in vitro is ALV - J virus gene, and it is nOt avian endogenous retrovir-us. The virus of NM0138 strain is the base for the further research..
