【摘要】 目的采用荧光、紫外光谱法研究在模拟人体生理条件下川芎嗪与人血清白蛋白(HSA)之间的相互作用。方法将川芎嗪对HSA进行荧光猝灭滴定,结合川芎嗪和HSA的紫外吸收图谱,确定川芎嗪对HSA的荧光猝灭机制;利用Scatchard模型和Frster非辐射能量转移理论得出川芎嗪和HSA的结合常数、结合位点数及结合距离;通过热力学常数确定川芎嗪和HSA的相互作用力类型。结果在温度为298K和303K时,川芎嗪与HSA的结合常数K分别为1.83×104L·mol-1和1.58×104L·mol-1,结合位点数n分别为1.06和1.05,结合距离r分别为3.74nm和3.55nm,相互作用力主要为静电作用力。结论通过川芎嗪与HSA结合的相关参数,阐明了川芎嗪和HSA的静态猝灭机制,建立了川芎嗪和HSA的体外结合模型。更多还原

【Abstract】 Objective To study the interaction between Tetramethylpyrazine and Human Serum Albumin under the simulative human physiological condition by fluorescence spectroscopy and ultraviolet spectroscopy.Methods The quenching mechanism of the fluorescence of Human Serum Albumin by Tetramethylpyrazine was studied by fluorescence quenching titration combined with ultraviolet absorbance spectrum.According to Scatchard’s mode and the theory of Fster’s non-radiation energy transfer mechanism,the binding parameters of Tetramethylpyrazine and Human Serum Albumin were determined,including the binding constant,the binding distance and the number of binding sites.Meanwhile,the type of interaction force was determined by the thermodynamic parameters.Results The binding parameters of Tetramethylpyrazine and Human Serum Albumin at 298K and 303K were as follows:The binding constants were 1.83×104L·mol-1 and 1.58×104L·mol-1,the number of binding sites were 1.06 and 1.05,the binding distance were 3.74 nm and 3.55 nm respectively,and the interaction was mainly driven by the electrostatic force.Conclusion The static quenching mechanism and the binding mode were demonstrated by the binding parameters of Tetramethylpyrazine and Human Serum Albumin.更多还原
