新型液相微萃取结合高效液相色谱法测定尿样中的利多卡因

【摘要】 应用新型液相微萃取与高效液相色谱联用技术快速分析尿样中的利多卡因。考察了萃取剂、体积、萃取时间、pH值及盐效应对液相微萃取的影响,建立了液相微萃取与高效液相色谱联用技术分析尿样中利多卡因的方法。在优化条件下,对浓度从0.02到2.0mg/L的利多卡因标准溶液系列萃取后进样分析。结果表明,利多卡因的色谱峰面积在所测浓度范围内与浓度呈良好的线性关系,回归方程为y=365.2x+4.962,相关系数为0.999,检出限为0.01mg/L,RSD小于4.4%(n=5)。样品回收率为106.1%。通过液相微萃取后,能有效地去除尿样中的干扰物质,获得了较高的选择性。该方法简单、方便、灵敏、消耗有机溶剂少,是尿样中利多卡因检测的一种有效方法。

【Abstract】 A method for determining lidocaine in human urine using a new single-drop LPME based on PCR tube in conjunction with HPLC-UV detection was developed.There are several parameters which determine the mass transfer such as the extraction solvent,drop and sample volumes,extraction time,pH and ionic strength,stirring rate.These parameters were chosen to optimize the performance in the current study.The method was validated with respect to precision,accuracy and specificity.The respective inter-day precision values were calculated to be below 4.4 %.The mean absolute recovery was 106.1%.The calibration graphs obtained by plotting the peak-height ratios against the concentrations of lidocaine added to the blank plasma were highly linear over the range 0.02-2.0mg/L,and the correlation coefficient for each calibration graph invariably exceeded 0.999.The mean regression equation is y=365.2x+4.962,where y is the concentration of lidocaine in human urine,and x is the peak area.The detection limits in urine were calculated to be 0.01mg/L.It was demonstrated that as long as the extraction procedure is consistently applied,quantitative analysis is performed accurately and reproducibly in human urine samples.
