β-1,3/1,6-葡聚糖硫酸酯的制备及其生物活性研究

【摘要】 目的制备β-1,3/1,6-葡聚糖硫酸酯并进行生物活性研究。方法以具有β-1,3/1,6-葡聚糖结构的水溶性灰树花多糖GF6F2为原料,以三氧化硫三甲胺盐(SO3·Me3N)为酯化试剂,合成了该多糖硫酸酯衍生物SGF6F2。结果所得产物酯化度为0.8,相对分子量为210 kDa。体外实验结果表明SGF6F2具有较好的抗凝活性及Src酪氨酸激酶抑制活性。结论灰树花多糖硫酸酯不仅具有一定的抗凝血活性,而且显示良好的抑制Src活性,为该糖类药物的开发提供了有用数据。
【Abstract】 Purpose To preparation ofβ-1,3/1,6-Glucans sulfate and study its bioactivities.Methods A water-soluble polysaccharide with structure ofβ-1,3/1,6-Glucans isolated from Grifola frondosa mycelium (GF6F2) was sulfated to SGF6F2 using sulfur trioxide trimethylamine complex (SO3·Me3N) as the reagent.Results Its degree of sulfation was found to be 0.8 and the relative molecular mass was 210 kDa.The bioassay in vitro results showed that SGF6F2 had a good anticoagulant activity and high Src inhibition ratio.Conclusion Theβ-1,3/1,6-Glucan sulfate not only showed good anticoagulation, but also inhibition to Src activity,these results offered useful data for polysaccharide based drug research..
