凹叶厚朴野生与栽培居群的遗传结构及遗传多样性

【摘要】 目的:研究凹叶厚各地方种群的遗传特性和亲缘关系,比较野生与栽培品遗传背景的差异,为栽培品与野生品的生药鉴定以及筛选和开发野生药材优质种质资源,提供重要参考。方法:利用 AFLP 技术对传统药用植物凹叶厚朴的7个野生居群和5个栽培居群的遗传结构和遗传多样性进行了分析。结果:凹叶厚朴遗传多样性指数 h=0.208,I=0.305,栽培居群保有野生居群95%的遗传变异,但差异并不显著。对于野生居群78.22%的变异分布于居群内,栽培居群则85.92%的变异分布于居群内。结论:凹叶厚朴目前仍保持适度较高的遗传变异,野生居群表现出比栽培居群更高的遗传多样性和更高的遗传分化,UPGMA 聚类不能将野生居群和栽培居群分开说明野生居群中的等位基因同样保留在栽培居群中。
【Abstract】 Objective:The goals of this study were to quantify the amount of genetic variation in wild and cultivated populations of M.officinalis subsp,biloba,compare the structure of genetic variation in wild and cultivated populations of M.officinalis subsp,biloba and establish if genetic diversity in wild populations was distinct from that in cultivated populations.Methods:The levels of genetic variation and genetic structure of seven wild populations and five domestic populations of M.officinalis subsp,biloba were estimated employing an AFLP methodology. Results At the population level,Nei’s measure of gene diversity(h),and Shannon’ s diversity index(Ⅰ),were 0.208 and 0.305,respectively.The cultivated populations maintained approximately 95% of the variation exhibited in wild populations.The analysis of genetic differentiation revealed that most of the AFLP diversity resided within populations both for the wild group(78.22%)and the cultivated group(85.92%).Conclusion The plant exhibited a relatively high level of intro-population genetic diversity.The cultivated populations of M.officinalis subsp,biloba maintain a subset of the total genetic diversity present in their wild progenitors and may play a crucial role in assuring the survival of the plant as native populations go extinct.Cluster analysis(UPGMA)did not discern among wild and cultivated populations,indicating that alleles from the wild population were maintained in the cultivated gene pool.
