木犀草素-7-O-β-D-葡萄糖苷的高效液相和分光光度分析方法研究

【摘要】 目的建立HPLC和UV两种方法测定以东紫苏为原料制得的产品中木犀草素-7-O-β-D-葡萄糖苷的含量。方法HPLC Agilent ODS(21mm×150mm,5μm);流动相:甲醇-1.5%冰醋酸水溶液(60:40),流速:0.3mL·min-1;检测波长:360nm;柱温:30℃;UV检测波长:360nm;结果HPLC和UV两种方法线性范围分别为0.3~0.7μg(r=0.9994)和0.004~0.014mg·mL-1(r=0.9999)。回收率分别为:99.5%(RSD=0.45%,n=5)和99.3%(RSD=1.2%,n=5)。结论两种方法无显著性差异,均可用于产品质量控制。
【Abstract】 OBJECTIVE To establish HPLC and spectrophotometric methods for determinatio- n of lutelin-7-O-β-D-glucoside in the product made from Elsholitzia bodinieri Vaniot. METHODS The separation was performed on a ODS(21 mm×150 mm, 5 μm)with methol-1.5% glacial acetic acid solution (60:40) as mobile phase at a flow rate of 0.3 mol·L-1 .The detection wavelength was 360 nm. RESULTS Both of HPLC and UV showed good linearity over the ranges of 0.3~0.7 μg (r=0.999 4), 0.004~0. 014 mg·mL-1(r=0.999 9) ,respectively .The average recoveries were 99. 5 %,99. 3 % wi- th corresponding RSDs of 0.45 % and 1.2%(n=5) ,respectively. CONCLUSION Results showed that no significant differences exist between the two methods. Both of the methods can be used for the determination of the contents of lutelin-7-O- β-D-glucoside for quality control of the product.
