NF-κB在小白菊内酯诱导人肝癌BEL-7402细胞凋亡中的作用

【摘要】 目的探讨NF-κB在小白菊内酯(PRT)诱导人肝癌BEL-7402细胞凋亡中的作用。方法体外培养人肝癌BEL-7402细胞株,用PRT处理,四唑盐比色试验(MTT法)、形态学观察法、DNA琼脂糖凝胶电泳方法检测细胞调亡;用免疫细胞化学法检测NF-κB活性。结果BEL-7402细胞在PRT作用下,细胞增殖被阻遏,并呈现剂量效应关系。同时诱导细胞发生凋亡。NF-κB在PRT处理BEL-7402细胞48h后,可抑制NF-κB活性。结论PRT可通过诱发BEL-7402细胞凋亡来阻遏BEL-7402的增殖,NF-κB可能参与PRT诱导人肝癌BEL-7402细胞凋亡中的信号转导。

【Abstract】 Objective To explore the role of nuclear factor Kappa B (NF-κB) in parthenolide (PRT) induced apoptosis in human hepatocellular carcinoma cell line BEL-7402. Methods Human hepatocellular carcinoma cell line was cultured in vitro and treated by parthenolide. Microculture terazolium assay (MTT), the study of the morphological changes, and gel electrophoresis assays were performed for the effects of PRT in various concentration on apoptosis in human hepatocellular carcinoma cells in vitro; immunohistochemistry was used as the determination method of NF-κB activity. Results PRT inhibits the proliferation of BEL-7402 cells in vitro and show dose-response relationship. Meanwhile, PRT can induce BEL-7402 cells to apoptpsis. Apoptosis cells demonstrated cell nuclear DNA and apoptotic peaks and apoptotic morphology; PRT treated BEL-7402 cells inviro after 48h, NF-κB activity was inhibited and dose-response relationship. Conclusion PRT could suppress the growth of human hepatocellular carcinoma cells via inducing them to apoptosis. PRT could rapidly inhibit activition and translocatin of NF-κB which could be one of signaling factor for inducing apoptosis.
