同时分离槐定碱等五种生物碱的实验研究

【摘要】 目的建立五种生物碱—槐定碱,苦参碱,氧化苦参碱,槐胺碱,槐果碱同时分离测定的高效液相色谱方法。方法色谱柱为ODS(3.9×150mm,5μm),流动相为0.01mol/L磷酸盐缓冲液(pH8.5)—甲醇(78∶22),流速1.0ml/min,检测波长为220nm。结果五种生物碱在上述色谱条件下达到完全的基线分离。槐定碱在10.21~204.10ug/ml(г=0.9999)、苦参碱在5.04~201.76ug/ml(г=0.9999)、氧化苦参碱在2.07~207.00ug/ml(г=0.9999)、槐胺碱在10.06~201.28ug/ml(г=0.9999)、槐果碱在5.04~201.49ug/ml(г=0.9999)的浓度范围内线性良好。结论本方法简便易行,可用于含该类生物碱的生药与制剂的分析。
【Abstract】 OBJECTIVE To establish a HPLC method for the simultaneous determination of Sophoridine , Matrine , Oxymatrine , Sophramine , Sophocarpine. METHOD Determination research was taken with ODS column(3.9×150mm,5um). And the mobile phase consisted of 0.01mol/L phosphate buffer solution(pH8.5)--methanol (78:22). The flow rate was 1.0ml/min. UV detection wavelength is 220nm. RESULTS A baseline separation was achieved for five alkaloids in determination. The established method shows good linearity. The linear range of Sophoridine is 10.21~204.10ug/ml(г=0.9999)、5.04~201.76ug/ml(г=0.9999) of Matrine、2.07~207.00ug/ml(г=0.9999) of Oxymatrine、 10.06~201.28ug/ml(г=0.9999) of Sophramine、5.04~201.49ug/ml(г=0.9999) of Sophocarpine. CONCLUSION The method appears easily to operate and suitable for the analysis of medicinal herbs and preparation which contains these alkaloids.
