刺参粗提物对神经细胞及肿瘤细胞生物活性的初步研究

【摘要】 [目的]研究刺参粗提物对体外培养的神经细胞及肿瘤细胞的生物学活性。[方法]刺参双酶解、醇沉,得到刺参粗提物。体外培养条件下将不同浓度的该粗提物添加到神经细胞、Bel-7402及C6两种肿瘤细胞的培养基中,通过荧光倒置显微镜观察刺参粗提物对神经细胞的作用,通过MTT法检测其对肿瘤细胞的影响。[结果]刺参粗提物可以促进神经细胞的生长分化。对Bel-7402肝癌细胞具有明显的增殖作用,以不加刺参粗提物的细胞为对照,当剂量为100、200、300、400μg/ml时,增殖率依次为41.3%、88.7%、148.91%、193.12%,而对C6细胞无明显影响。[结论]刺参粗提物对于神经细胞的生长分化具有促进作用,而对于肿瘤细胞则具有不同程度的增殖作用。

【Abstract】 OBJECTIVE To investigate the bioactivity of the extraction of Stichopus japonicus on nerve cell and tumor cell. METHODS We used pepsin and trypsin to hydrolyze protein, and precipitated what we want by ethanol. Then we added what we extacted from Stichopus japonicus into the nutrient medium of nerve cell and tumor cell respectively. After some time,we observed the state of nerve cells by phase contrast fluorescence microscopy, and use MTT to assay the effect of Stichopus japonicus extraction on tumor cells. RESULTS Our results showed that Stichopus japonicus extraction can encourage the growth and differentiation of nerve cell. With respect to Bel-7402 liver tumor cells, the growth rate was 41.3%、88.7%、148.91%、193.12% when the amount of Stichopus japonicus extraction added into the nutrient medium was 100、200、300、400μg/mlrespectively. But Stichopus japonicus extraction has little effect on C6 nerve neurospongioma cells. CONCLUSION We deemed that Stichopus japonicus extraction can encourage the growth and differentiation of nerve cell. And it has different proliferation effect on different tumor cells.
