丹参素大鼠在体肠吸收动力学的研究

【摘要】 目的:考察丹参素在大鼠各肠段的吸收动力学特征及不同药物浓度对其的影响。方法:采用大鼠在体肠吸收实验方法,用HPLC方法对循环液中的丹参素进行分析。结果:在2.0~16.0mg·L-1的浓度范围内丹参素的吸收量与浓度成线性关系,Ka值基本保持不变;各肠段的吸收速率有显著性差异,十二指肠、空肠、回肠、结肠的Ka值分别为0.09034±0.02732,0.04887±0.01301,0.00961±0.00246,0.01157±0.00249h-1。结论:丹参素在肠道的吸收呈现一级动力学过程,且吸收机制为被动扩散。

【Abstract】 OBJECTIVE: To investigate the absorption kinetics of danshensu at different intestine segments in rats and the influence of the drug concentration on the absorption kinetics. MRTHODS: The intestine in rats was cannulated for in situ recirculation. Danshensu concentration in the flux was measured by the reversed phase HPLC. RESULTS: When the concentration of danshensu was raised from 2.0 to 16.0 mg·L-1, the uptake of danshensu increased linearly, whereas the permeability coefficient kept at an equilibrium level. The permeability coefficient of danshensu at duodenum, jejunum, ileum and colon were 0.09034±0.02732, 0.04887±0.01301, 0.00961±0.00246, 0.01157±0.00249h-1, respectively and which were different notability. CONCLUSION: The absorption of danshensu in rat’s intestine was a first-order process with the passive diffusion mechanism.
