胸腺五肽溶液稳定性考察

【摘要】 目的考察胸腺五肽在不同pH、缓冲盐及温度条件下的稳定性。方法建立了胸腺五肽的HPLC检测方法,采用C18柱,流动相50mmol/L磷酸二氢钾(pH7.0)-甲醇(85:15);流速1ml/min;检测波长275nm。通过长期和加速试验,检测胸腺五肽在一定条件下的含量变化。结果37℃和60℃时,胸腺五肽在两种缓冲溶液中的稳定性随着pH值下降而下降。在相同pH条件下,胸腺五肽在磷酸缓冲液中降解较少。结论pH值对胸腺五肽溶液稳定性有较大影响,与在柠檬酸缓冲液中相比,胸腺五肽在磷酸缓冲液中更加稳定。

【Abstract】 OBJECTIVE To study the stability of thymopentin under different pH, buffer and temperature. METHODS A HPLC method was established for examination of Thymopentin, a C18 was used with the mobile phase of 50mmol/L Potassium dihydrogen phosphate (pH7.0)-methanol (85:15), at the detection wavelength of 275nm, the flow rate was 1ml/min. Through long-term and accelerated experiments, the change of the content of thymopentin was examed. RESULTS The stability of thymopentin degrades as the pH degrades under 37 or 60℃. At the same pH, thmopentin degrades less in the phosphate buffer than in the citrate buffer solution. CONCLUSION The results showed that the pH has a great effect on the stability of thymopentin solution, when the pH is 3.0, thymopentin degrade most within one month. Compared with the citrate buffer solution, thymopentin solution is more stable in the phosphate buffer.
