P2受体在食管癌细胞株和组织中的表达及ATP对其表达的影响

【摘要】 目的:通过测定P2嘌呤和嘧啶受体在食管癌细胞株和组织中的表达及ATP对其表达的影响,探讨外源性核苷酸及其受体在肿瘤药理学与治疗学中的作用,为肿瘤治疗寻找新的靶点。方法:采用逆转录-聚合酶链反应(RT-PCR)技术分别对 TE-1、TE-10、TE-11、TE-13和TTn食管鳞癌细胞株以及食管癌患者手术切除癌及远癌正常组织中P2X7、P2Y1和P2Y2受体的表达及ATP对其表达的影响进行了研究。结果:P2Y1受体mRNA在5株食管癌细胞株和组织中全部表达,P2Y2受体 mRNA在5株食管癌细胞株、5例食管正常组织和4例癌组织中表达,P2X7受体mRNA仅在TTn食管癌细胞株弱表达,7 例患者的食管正常组织和癌组织中均不表达P2X7受体。ATP处理TE—13细胞48h后,均可上调P2Y1和P2Y2受体mRNA 的表达。0.1,0.3,1 mmol·L-1 ATP处理后,细胞P2Y1受体表达的百分比分别比对照组增加了:43.2%,23.09%和61.32%。 1 mmol·L-1 ATP处理组的P2Y2受体表达的百分率比对照组增加了:14.92%。结论:人食管癌组织中普遍存在P2Y1和P2Y2 受体,P2Y1和P2Y2受体有可能在食管癌的药理治疗中发挥重要的作用,有望成为今后肿瘤治疗的新靶点。

【Abstract】 AIM To study the distribution of P2 receptor in esophageal carcinoma cells and tissues, as well as the changes of expression induced by ATP. METHODS RT-PCR were used to determine the expression of P2X7, P2Y1, P2Y2 receptors mRNA on several tumor cell lines, included human squamous esophageal carcinoma cells TE-13, TE-1, TE-10, TE-11 and TTn, and tissues from 7 esophageal cancer patients. RESULTS All of the five esophageal cancer cell lines expressed the P2Y1 and P2Y2 receptors. Of the seven esophageal cancer patients, all the normal and cancer tissues expressed P2Y1 receptor; P2Y2 receptors were expressed in 5 cases of normal tissues and 4 cases of cancer tissues. Only one of the five cells, TTn cells expressed P2X7 weakly. Neither the normal nor cancer tissues of 7 esophageal cancer patients expressed it. Constitutive P2Y1 and P2Y2 receptor mRNA expression was obviously increased by treatment with ATP for 48 h at concentrations of 0.1, 0.3, 1 mmol·L-1 by 43.2%, 23.09% and 61.32% for P2Y1 receptor mRNA (P<0.05), and by 14.92% for P2Y2 receptor mRNA (1 mmol·L-1).CONCLUSION These data demonstrated that all the esophageal cancer cell lines and tissues tested expressed the P2Y1 and P2Y2 receptors, indicating that a pharmacological role for P2Y1 and P2Y2 receptors in the esophageal cancer.
