【摘要】 目的观察保存前白细胞过滤对单采血小板保存过程中TGF-β1含量的影响。方法应用细胞分离机单采血小板,然后经白细胞过滤,并于标准条件下保存5d,同时设自身对照。间隔取样用ELISA法检测TGF-β1含量。结果对照组在保存1d后,TGF-β1含量与保存前比较即有显著升高(P<0.01),过滤组在保存3d后TGF-β1含量与保存前比较有显著升高(P<0.01),且在保存期中其含量要显著低于对照组(P<0.01)。结论保存前过滤白细胞能有减少TGF-β1的产生,从而有效地预防血小板输注所引起的免疫抑制。更多还原

【Abstract】 Objective:To observe the effects of prestorage leucocyte filtration on the transforming growth factor-β1 (TGF-β1) levels in stored apheresis platelets.Methods:Apheresis platelet were prepared by cell separator,then filtrated by leucocyte filter and stored at standard condition for 5 days,self-control was established,contents of TGF-β1 were measured by ELISA at various storage time. Result:The TGF-β1 levels in apheresis platelets of control group after 1 day storage were obviously higher than that of prestorage(P<0.01).In filtration group,the TGF-β1 levels in apheresis platelets after 3 day storage were obviously higher than that of prestorage(P<0.01),and the contents of TGF-β1 were much lower than that of control group during storage(P<0.01).Conclusion:Prestorage leucocyte filtration can reduce the production of TGF-β1 and could be effectively prevent the immunosuppressive effect of platlet transfusion.更多还原
