小鼠胚胎心脏发育形态学研究与p300和CBP在胎心中的表达

【摘要】 目的:观察小鼠胚胎心脏发育过程及组蛋白乙酰化酶 p300和 CREB 结合蛋白 (CBP)在小鼠胚胎心脏上的时空表达规律,为进一步探明其与先天性心脏病的发生之间的关系奠定基础。方法:选取胎龄7.5-18d 和出生1d 小鼠正常心脏36例,采用免疫组化法观察 p300和 CBP 在小鼠胎心发育各个时期的表达分布及变化规律。结果:小鼠胚胎7.5天出现心脏新月,此期 p300强表达而 CBP 不表达。E8.5~E9.5出现原始心管并环化,E10.5房室管形成并出现心内膜垫和突入心腔的小梁,E11.5~E13.5房间隔形成,E11.5~E14.5室间隔形成, E12.5~E15.5房室瓣形成,E11.5~E16.5心肌细胞增殖使心室壁致密变厚,E16.5~生后1d 心肌细胞增大使心脏体积变大。CBP 与同期同区域的 p300相比,有相对低的阳性细胞表达率,二者差异有统计学意义(P<0.05)。结论:p300和 CBP 都与胚胎心脏发育密切相关,他们在发育心脏中的表达时空性不同,p300较 CBP 表达更早更广,可能也更强,p300对整个胚胎心脏发育尤其是发育早期的心脏新月和中期的室间隔形成作用更大。

【Abstract】 Objective:This study was to investigate the progress of mouse heart development and the spatiotemporal expressions of histone acetyltransferases p300 and CREB binding protein (CBP) in the developing mouse heart.Which,lay the foundations to further clarify the relations between them and congenital heart diseases.Methods:36 samples from normal mouse fetal heartsin embryonic days 7.5-18 and postnatal 1 day were included in this study. Immunohistochemistry was used to detect the distributions and expression changes during every stages of mouse cardiac crescent at E7.5.At E8.5-E9.5,primary heart tube formed and looped,At El0.5,atrioventricular canal formed as well as atrioventricular endocardiac cushion and protrusive trabeculae appeared.Interatria septum formed at E11.5-El 3.5 and interventricular septum formed at E11.5-E14.5 and atrioventricular valve formed at E12.5-E15.5.Myocardial cell multiplication caused the ventricle to compact and thicken at E11.5-E 16.5 while myocardial cell increase caused the heart to greaten at low rate of positive cells expressed,the difference is statistically significant (P<0.05).Conclusion:p300 and CBP was related to embryo heart development,while the spatiotemporal expressions of them during mouse embryo heart evelopment were different.The expression of p300 was earlier and broader as well as presumptively stronger,p300 may play greater role in embryo heart development especially for cardiac crescent and interventricular septum formation than CBP
