芦荟中某些天然抗氧化产物的核磁共振表征与特性分析

【摘要】 芦荟系百合科(Liliaceae)芦荟属(Aloe)多年生常绿肉质草本植物。现代药理实验证明:该属植物中的有效部位或有效成分具有抗癌、消炎、杀菌、抗病毒、杀虫解热、保肝及增强免疫等功效。芦荟在医药、保健品、日用化工等诸多方面存在着独特的潜在应用价值,引起广大科研工作者对其活性成分研究的兴趣。近年来本工作组已从中药芦荟(Aloe vera)中分离出十多种含酚羟基的天然化合物,它们包括蒽醌、色酮、双氢异香豆素、香豆素与芦荟甙类衍生物等等。其中五种化合物属自然界首次发现。这些天然化合物分子结构的鉴定与表征主要通过 HR-MS 与2D-NMR(H-H COSY、 HMQC、HMBC 和 J-resolved)技术,并辅助单晶 X-ray 分析、UV、IR 与 CD 等等。在此基础上,作者又进一步对比研究了这些化合物的抗氧化性能。并针对不同类型的抗氧化分子反应模型从理论上对它们的抗氧化作用进行了系统地总结与分析,探索了这些化合物抗氧化能力与分子结构间的构效关系。

【Abstract】 Aloe is well known for its wide uses in health foods,medicines and cosmetics, which attracts the great interests of many researchers.Biological studies revealed that the leaves of this species possess purgative,analgesic,antipyretic,anticancer, antimicrobial and immunological activities.In resent years our group have already isolated and purified more than ten kinds of natural polyphenol products,such as: anthraquinones,chromones,dihydroisocoumarins,coumarins,aloin derivatives and etc.Of the above natural products,five were first discovered.The structures of these compounds were determined by high-resolution negative fast atom bombardment mass spectrometry(HR-FAB-MS)and NMR spectroscopy. Complete 1H and 13C NMR chemical assignments of these compounds were achieved using one-and two-dimensional NMR techniques,including 1H-1H COSY,HMQC and HMBC experiments.The spectral properties(MS,IR,UV and CD)and single-crystal X-ray analysis are also presented.Finally,we have performed some anti-oxidative experiments of these compounds,and then systematically analysed the correlation between their antioxidant activity and the molecular structures by means of molecular modeling.
