环丙沙星与β-环糊精固体配合物的合成和核磁共振研究

【摘要】 <正>环糊精是由 D-吡喃葡萄糖基通过α-(1→4)糖苷键组成,最主要的产品有三种:α,β和γ-CD,分别含,7,8个葡萄糖基,结构相似,只是形成的空腔的直径和高度不同。环糊精的特殊的外亲水,内疏水的分子结构使得它能与多种疏水性化合物形成主-客包结配合物,从而改善了客体分子的物理,化学,生物学性质,最重要的是对生物体毒性小,是很有前途的药物载体材料。固体环糊精在医药工业有重要的用途,并且也广泛应用于分析化学方面。
【Abstract】 Cyclodextrins(CDs) are polysaccharides made up of six to eight D-glucose monomers connected at the 1 and 4 carbon atoms.They have the property of forming inclusion complex with various guests molecules with suitable polarity and dimension because of their special molecular structure-hydrophobic internal cavity and hydrophilic external surface.The interaction of ciprofloxacin with β-CD has been studied by several analytical techniques,including 1H- NMR,13C-NMR,2DNMR,fluorescence spectra,infrared spectroscopy,thermal analyses and scanning electron microscope etc.In this paper,solid inclusion complex of ciprofloxacin with β- CD was synthesized by the coprecipitation method.In addition,the characterization of the inclusion complex has been proved by fluorimetry,infrared spectra,differential scanning, calorimetry and 1D,2D NMR.The experimental results confirmed the existence of 1:1 inclusion complex of ciprofloxacin with β-CD,and the formation constant of the inclusion complex is 5.0× 102mol/l.A spatial configuration of inclusion compound has been proposed based on these data.
