用固相萃取-核磁共振谱法对光活布洛芬在大鼠体内代谢产物的研究

【摘要】 <正>目前,高分辨核磁氢谱已经有可能从尿、血浆等生物样品中直接检测和鉴定药物及其代谢产物(1,2)。固相萃取技术可以把大部分内源性物质除去,采用梯度洗脱的方法可以把代谢混合物分成若干部分,以利于药物代谢产物的结构解析。
【Abstract】 Ibuprofen(2-[4’-isobutylphenyl]-propionic acid,IBP) is an important nonsteroidal anti- inflammatory drugs with antipyretic activities.We study the metabolites ofchiral IBP in rat urine by SEP-PAK column and NMR.A stepwise gradient of methanol and water was applied and fractions were scanned by proton NMR.The main metabolites are 2’- hydroxy-IBP、2’-hydroxy-IBP-glucuronide、2’-carboxy-IBP and IBP-glucuronide in the spectra of(±)-IBP which shows no differece with that by Wilson I D.And we didn’t discover taurine-IBP and glycine-IBP which are derived from the transformation from (- )-IBP to (+)-IBP.There is no main difference in the total spectrums of racemic and chiral IBP.But in the spectra of 10% and 60% fractions of R-IBP,some peaks appeared.It demonstrates that R-IBP did not inverted to S-IBP entirely,and it has some new metabolites.
