ω-羟基烷酸甲酯的脂肪酶催化合成环十五内酯的研究

【摘要】 采用己筛选到的假丝酵母菌株GXU0 8所产脂肪酶来催化底物15 -羟基十五烷酸甲酯关环合成环十五内酯,考察了脂肪酶活力、反应时间、温度、pH值、有机溶剂极性等对合成转化率的影响。结果表明,在混合羟基脂肪酸甲酯中,GXU0 8具有很好的专一性,未发现环九内酯,环十三内酯及分子间聚合的二聚体等产物。底物于4 0℃、180rpm、pH =6 0 ,脂肪酶活力12 4 5U的条件下在环己烷中反应72h ,转化率为17%。
【Abstract】 One kind of screened lipase from Candida sp GXU08 was used as a catalyst to synthesize Cyclopentadecanolide Methyl 15-Hydroxy-pentadecanate was used as the substrate Such effects as activity of lipase, time of reaction,temperature, PH, dielectric constant of organic solvent etc on the conversion have been studied The results indicate that GXU08 has good specificity and no cyclononalide,cyclotridecanolide or other polymers have been found Under the optimum reaction condition: 40℃,180rpm,pH=6 0,activity of lipase 1245U , 72h ,the conversion is 17% in cyclohexane
