不同添加物对猪卵母细胞体外成熟的影响

【摘要】 本实验就PMSG、hCG、E2及rpGH对猪卵母细胞体外成熟的影响进行了研究,并比较了BSA、NCS、FCS和L-cysteine在成熟中的作用。结果表明,hCG和PMSG联合使用的促成熟作用显著高于PMSG单独使用(31.9%、17.5%,P<0.05);E2在成熟中的作用不显著(35.0%、31.9%,P>0.05)。成熟中NCS的促成熟效果与FCS无显著差异(P>.05),但两者均显著高于BSA和BSA+FSC(38.4%、30.0%、20.0%、27.3%,P<0.05)。添加6mg/mL-cys和10μg/L rpGH后,体外成熟率为30.4%和50.8%,显著高于其它浓度组(P<0.05,P<0.01)。实验表明,在猪卵母细胞IVM中添加10IU/L hCG、PMSG,0.6mmol/L L-cysteine和10μg/L rpGH有助于卵母细胞成熟;同时添加10%FCS或10%NSC比较适宜。
【Abstract】 The effects of PMSG, hCG, E2, rpGH, BSA, NCS, PCS and L-cysteine on the IVM of porcine oocytes were studied in this paper. The results demonstrated the rate of matured oocytes with mixture of hCG and PMSG(31.9%)was significantly higher than that of PMSG (17.5%)(P<0.05). Supplemented NSC, PCS, BSA and mixture of BSA and PCS to the IVM system, the rates were 38.4% , 30.0% , 20.0% and 273% respectively. Both NSC and PCS were significantly higher than others(P<0.05). The rates of IVM supplemented with 6 mg/mLL-cys (30.4%) or 10 μg/L rpGH(50.8%) were significantly higher than others. The study indicated the IVM system added 10IU/L hCG, 10IU/L PMSG, 0.6 mmol/L L-cysteine, 10 u g/L rpGH , 10% PCS or 10% NSC was useful to the mature of porcine oocytes in vitro.
