转基因猪研究进展
Advances on the Reseach of Transgenic Swine

【摘要】 本文综述了湖北省农科院生物技术研究所有关转基因猪研究的主要进展。先后将POMT-PGH,PSV40-PGH,POMT-PGH-Micro satellite DNA,PMHR,hDAF,pBHSA,pSHSA等七种不同结构的外源基因,注入4027枚猪的受精卵原核,获得原代表达的转基因猪83头,转基因的效率在0.9%~3.4%之间,平均2.2%。通过几个主要技术环节的改进,提高了转基因的效率;建立了外源基因整合的定性检测(PCR,Dot blot和Southern blot)和定位检测-染色体原位杂交技术;进行了相应外源基因的蛋白质表达检测:研究了转基因猪的遗传规律。
【Abstract】 This paper summarized the advances on transgenic swine in Biotech Institute of Hubei Academy of Agriculture Science. The Seven individual foreign genes with different construction, including POMT-PGH, PSV40-PGH, POMT-PGH-Micro satellite DNA, PMHR, hDAF, pBHSA, pSHSA and so on, were microinjected into 4027 embryos. 83 founders were produced. The efficiency was percent 0.9 to 3.4 and percent 2.2 for average. Through improving main technological process, we, arose the efficiency of transgenic swine and established the technology of hybrid in situ of chromosome and detection foreign gene quantity by PCR , Dot blot, Southern blot and Northern blot for expression protein to investigate the genetic regulation of transgenic swine.
