胰岛素和肾上腺素对前体脂肪细胞增殖与分化的调控

【摘要】 以生长发育正常、健康状况良好的20日龄雄性SD大鼠为实验动物,采用脂肪细胞离体培养、MTT比色、流式细胞仪分析、GPDH活性检测等技术,系统地研究了胰岛素(INS)、肾上腺素对大鼠前体脂肪细胞增殖与分化的调控。结果表明:INS可促进大鼠前体脂肪细胞的增殖,而肾上腺素则抑制前体脂肪细胞的增殖,其中25μU/mL INS(生理浓度)处理的前体脂肪细胞增殖效果好于0.1U/mL INS(药理浓度),10-5M肾上腺素(药理浓度)抑制前体脂肪细胞增殖的效果高于10-7M肾上腺素(生理浓度);INS可降低G1期细胞比例,提高PrI期(细胞增殖指数)的细胞比例,肾上腺素提高G1期细胞比例,降低PrI期细胞比例,其中25μU/mL INS促进增殖的效果最好,10-5M肾上腺素抑制增殖的程度最强;INS可使前体脂肪细胞中的GPDH活性极显著地高于对照组(P<0.01),肾上腺素处理的前体脂肪细胞中GPDH活性极显著地低于对照组(P<0.01),其中25μU/ml INS升高细胞中GPDH活性的幅度最大(约4倍),10-5M肾上腺素降低细胞中GPDH活性的幅度最大(约3倍)。

【Abstract】 By cell culture, MTT method, flow cytometer, GPDH activity detection, normal and healthy male SD mice with 20 days age were used to systematically explore the regulation of INS and adrenal hormone on the proliferation and differentiation of mice preadipocytes. Following results were obtained: INS enhances, however, adrenal hormone depressed the proliferation and differentiation of mice preadipocytes. Proliferation is more effective when the preadipocytes were treated with INS at the concentration of 25 μU/ml (physical concentration) than 0.1 U/ml (pharmaceutical concentration), and 10-5 M (pharmaceutical concentration)of adrenal hormone has a better depression on preadipocytes’ proliferation than 10-7 M (physical concentration). INS can decrease the cell ration in Gl period, increase it in PrI period, especially when INS is at the concentration of 25 μU/ml, but increases the cell ration in Gl period, decreases it in PrI period, especially when adrenal hormone is at the concentration of 10-5 M. INS can make the GPDH activities remarkably higher than the control, and GPDH activities of preadipocytes treated with adrenal hormone are much lower than the control. At the concentration of 25 μU/ml, INS can improve 4 times of the GPDH activities, adrenal hormone can depress them 3 times at the concentration of 10-5 M.
