脂肪型和瘦肉型猪H-FABP基因分子标记研究

【摘要】 本试验利用PCR-RFLP(HinfⅠ、HaeⅢ和MspⅠ3种限制性内切酶)分子标记技术,检测了杜洛克、长白、大白、内江猪、荣昌猪、汉江黑猪、汉白猪、八眉猪和野猪共计265头猪心脏脂肪酸结合蛋白基因5’-上游区和第二内含子区的遗传变异。结果表明,在HinfⅠ-RFLP位点上,上述猪种和野猪均存在多态性,等位基因H的频率分别为0.7500,0.7188,0.9167,0.3333,0.1250,0.6909,0.1167,0.8500和0.9375;除汉江黑猪(P<0.05)和野猪(P<0.01)外,其余的猪种基因频率和基因型频率都处于Hardy-Weinberg平衡状态(P>0.05);大白、八眉、汉江黑猪、汉白猪和野猪表现为低度多态(PIC<0.25),杜洛克、长白猪、内江猪和荣昌猪为中度多态性(0.25<PIC<0.5);而在HaeⅢ-RFLP和MspⅠ-RFLP位点上,仅内江猪、荣昌猪、汉江黑猪和八眉猪为单态。

【Abstract】 In this experiment, genetic variations of 5’-upstream region and the second intron of H-FABP gene in 265 pig from Duroc, Landrance, Largewhite, Neijiang pig, Rongchang pig, Hanjiang black pig, Hanzhong white pig, Bamei pig and wild pig were detected by PCR-RFLP molecular marker with HinfⅠ, HaeⅢ and MspⅠ. The results showed that polymorphisms for HinfⅠ-RFLP in above pig breeds were detected and frequencies of allele H were 0.7500, 0.7188, 0.9167, 0.3333, 0.1250, 0.6909, 0.1167, 0.8500 and 0.9375 respectively; except in Hanjiang black pig (P<0.05) and wild pig(P<0.01), frequencies of gene and genotype in other breeds were in equilibrium of Hardy-Weinberg (P>0.05); Large white, Bamei pig, Hanjiang black pig, Hanzhong white pig and wild pig presented low polymorphisms (P/C<0.25), while other breeds have mediated polymorphisms (0.25<PIC<0.5) . Polymorphisms of H-FABP gene for the HaeⅢ-RFLP and MspI-RFLP were not detected in only 4 Chinese local pig breeds (Neijiang pig, Rongchang pig, Hanjiang black pig and Bamei pig Bamei pig) among tested breeds.
