BMPR-IB和BMP15基因作为中国美利奴多胎品系和湖羊侯选基因的研究

【摘要】 以控制Booroola Merino羊多胎性能的BMPR-IB基因,以及影响Invedale和Hanna羊排卵数的BMP15基因作为候选基因,从分子水平上对中国美利奴多胎品系和湖羊进行多胎机制的研究,分析其单核苷酸突变位点(SNPs)。并通过大规模的群体检测统计推测其遗传效应。实验结果表明:中国美利奴多胎群体和湖羊在BMPR-IB基因的相应位置上发生了与Booroola Merino羊相同的突变位点(A746G),在这两个品种(系)中出现了3种基因型(BB、B+、++),该基因的BB和B+基因型在中国美利奴多胎品系和湖羊群体内为优势基因型。而且中国美利奴多胎群体BB和B+基因型母羊比++型分别多排2.44(P<0.01)和1.94(P<0.01)枚卵,多产0.891(P<0.01)和0.887(P<0.01)只羔;湖羊BB和B+基因型母羊比++型分别多产0.875(P<0.01)和0.800(P<0.01)只羔。BB和B+基因型母羊的排卵数和产羔数明显高于++基因型。BMPR-IB基因在中国美利奴多胎品系和湖羊群体基因型的分布和各基因型与排卵数和产羔数的高度相关的研究结果表明:BMPR-IB基因是影响中国美利奴多胎品系和湖羊多胎性能的主效基因,可以用于对绵羊产羔数的选择。BMP15基因在中国美利奴多胎品系和湖羊群体中不存在V31D(T896A)和Q23Ter(C871T)突变,这说明中国美利奴多胎品系和湖羊的多胎遗传机制与Invedale和Hanna羊不同,排除了BMP15基因突变影响中国美利奴多胎品系和湖羊排卵数和产羔数的可能性。

【Abstract】 BMPR-IB gene which controls the fecundity of Booroola Merino and BMP15 gene Which affects the ovulation of Inverdale and Hanna were studies as candidate genes on fecundity of China Merino fecundity sheep and Hu-Yang, and their mutation and genetic effects were analyzed. The results showed there was a same mutation in BMPR-IB gene (A746G)of China Merino fecundity sheep and Hu-Yang. There were three genotypes (BB, B+, ++), the BB and B+ mutation genotype was superior in prolific China merino fecundity sheep and HU-YANG. Further more, the ewes with genotype BB and B+ had 2.44 (P<0.01) and 1.94 (P<0.01) ovulations, had 0.89 (P<0.01) and 0.887 (P<0.01) lambs more than those with genotype in the China merino fecundity sheep, respectively; in the Prolific HU-YANG , the ewes with genotype BB and B+ had 0.875 (P<0.01) and 0.800 (P<0.01) lambs more than those with genotype. The ewes with genotype BB or B+ had a significant higher ovulation and litter sizes than ++ genotype ewes , and genotype had a directly relativity with ovulation and litter sizes. It could be inferred that BMPR-IB gene was the major gene that controls the high prolificacy of China Merino fecundity sheep and Hu-Yang, it could be used as the molecular genetic marker to select litter size in sheep. While there was not mutation of V31D (T896A) or Q23Ter (C871T) in BMP15 gene of China Merino fecundity sheep and Hu-Yang, it showed that the fecundity mechanism of China Merino fecundity sheep and Hu-Yang was different from that of Invedale and Hanna, then it ruled out the possibility that the ovulation of China Merino fecundity sheep and Hu-Yang was affected by the mutation of BMP15 gene.
