香蕉横切薄层培养及其植株再生
Efficient Plant Regeneration of Banana (Musa spp.)using Thin-cell-layer Culture

【摘要】 以天宝蕉吸芽茎尖培养产生的分生芽为外植体,通过薄层培养和愈伤组织的诱导,为香蕉的转基因再生系统研究提供材料。研究发现横切薄层切片的芽再生率高于纵切,而愈伤组织诱导的最佳培养基配方为MS+2,4-D6+KT0.5+AgNO35mg/L+Su3%+Ag0.9%。香蕉芽增殖的最佳培养条件为30℃黑暗培养。
【Abstract】 A protocol was developed using banana (Musa spp.) shoot tip as explants,through thin-cell-layer culture and callus induction to produce the maximum numbers of shoots,which was the material for banana transformation and regeneration. It shows that the shootsregeneration frequency of transverse thin-cell-layer culture was higher than longitudinalthin-cell-layer culture. The best medium for callus induction was MS-based mediumsupplemented with 6mg/1 2,4-D, 0.5mg/1 KT, 3% sugar and 0.9% agar. The best condition forbanana multiplication was cultured in dark at 30℃.
