多效唑在生姜组织培养中的应用研究

【摘要】 本实验以MS为基本培养基,兴国九山姜为材料,研究多效唑(PP333)在生姜试管苗快繁、壮苗、移栽、试管姜诱导及种质保存中的应用。结果表明:①在生姜试管苗扩繁培养基中,附加1.5mg/L的多效唑可使试管苗增殖与壮苗同步进行,得到的试管苗矮壮,根系发达,大大提高移栽成活率;②2.5mg/L多效唑配合0.2mg/L 6-BA,0.5 mg/L NAA,2.0mg/L磷酸钙和8%的蔗糖使用,有利于提高试管姜的诱导率和大小;③在MS+8.0~10.0mg/L多效唑的培养基中,生姜试管苗的种质保存期可延长至4~5个月。
【Abstract】 In the experiment, using MS medium as basic medium and Jiushan-ginger asmaterial, the effects of PP333 on multiplication, strengthening, transplant, induction ofmicro-ginger and conservation in vitro are studied. The main results are showed in thefollowings. (1) Adding 1.5 mg/L PP333 to multiplication medium, strengthening can be finishedwith multiplication of seedling in vitro. The seedlings in vitro get lower and stronger, rootsdeveloped. The survival rate of transplant is highly increased. (2) The micro-ginger formationcan be promoted in the medium with 2.0 mg/L PP333 +0.2 mg/L 6-BA+0.5 mg/L NAA+ 8% sucrose+2.0 mg/L Ca3(PO4)2. The rate of induction is 180%, and the micro-gingers are bigger.(3) Adding 8~10 mg/L PP333 to MS medium, the period of conservation of ginger in vitro atnormal temperature can be prolonged to 4~5 months.
