怀山药脱毒快繁技术研究

【摘要】 本文首次对怀山药感染病毒的种类和症状、脱毒苗脱毒、快繁、生根、移栽以及大田种植和产量高低进行了研究。结果表明:(1)怀山药感染病毒后的症状为叶色失绿、花叶、畸形;电镜法和PCR方法证明怀山药感染的病毒为马铃Y病毒和马铃薯卷叶病毒,它是造成怀山药产量下降和品质退化的主要原因;(2)怀山药茎尖在供试培养基上,可脱分化形成愈伤组织,然后再分化形成再生植株,也可直接形成再生植株;(3)茎尖培养可以脱除病毒,但脱毒效果与茎尖大小有关;(4)KT2.0+NAA0.02有利于脱毒苗的繁殖;(5)PP3330.5-1有利于脱毒苗的壮苗和生根,处理的脱毒苗生长健壮、叶色浓绿、移栽成活率高;(6)脱毒苗移栽大田后生长势强、健壮、抗逆性强,产量分别比非脱毒苗高67.7%和47.1%。

【Abstract】 In this paper, the categories and symptoms infected by virus, virus-freeTechnology, Microprogation, outgrowing roots, transplanting culturing in fields and yield areresearched. The results are as follows: (1) The symptoms of being infected by virus are leavesloosing green, flower leaves are abnormality; electron microscope technology and PCR showsthat the infected viruses are potato Y virus and potato rolled-leaf virus, which are the mainreason of yield decreasing and quality degenerating. (2) In the supplied medium, Dioscoreaopposita stem nodes can first induced into callus, then polarizing into regenerating plants. (3)Stem culture can free virus, and the effect is combined with the size of stem nodes. (4) KT2.0+NAA0.02 is beneficial to the propagation of Virus-free plants. (5) PP333 0.5-1 is beneficial tostrengthen plants and germinate roots, and the virus-free plants grow strongly, haveheavily-green leaves and have high survival rate. (6) The virus-free plants have strong growingtendency and high tolerance, and the yields increased 67.7% and 47.1% respectively.
