小麦黄花叶病转基因研究

【摘要】 筛选出适宜小麦转化的优良基因型扬麦158和扬麦10号。以扬麦158的胚愈伤组织为受体,利用基因枪共转化法将小麦黄花叶病毒复制酶基因(WYMV-Nib8)转入了小麦。T0代经PCR扩增分析,获得了14株含WYMV-Nib8基因的转化植株,T1、T2代跟踪分子检测和T2、T3代田间抗性鉴定结果表明,获得了4个抗WYMV的转基因株系,同时利用PCR技术筛选到只含功能基因而没有标记基因的扬麦158转基因小麦。

【Abstract】 Two wheat genotypes with high tissue culture ability, Yangmai 158 andYangmai 10, were screened from ten commercial varieties for the followingtransformation. Then, the seven days immature embryo calli of Yangmai 158 used asreceptor of Nib8 target gene, which was isolated from wheat yellow mosaic virus, werebombardmented by biolistic particle. A lot of resistant plantlets to 3∽5 mg/l Biolophoswere achieved on the selection medium. By PCR analysis in the T0 generation, fourteen transgenic plants were identified. Four transgenic wheat lines resistant to the diseasestrongly were obtained finally combining the molecular test of T1, T2 and WYMV field testof T2.,T3 Excision of bar gene from transgenic wheat through the methods of PCR analysisand shoes the transgenic wheat which only have target gene without selectable markergene.
