水稻苯达松敏感致死基因的电子杂交定位及基因预测

【摘要】 水稻苯达松敏感致死基因(ben)在杂交水稻制种生产中具有广阔的应用前景。本研究根据中国水稻全基因组框架序列中的跨叠克隆Contig10968,成功地将与Ben/ben共分离的RAPD标记OPG18/972和OPG18/943步移延伸至8138bp和8103bp。利用生物信息学技术,通过电子杂交将OPG18/972和OPG18/943及步移后的序列定位于第2染色体150.5cM处,进一步将Ben/ben初步定位于第2染色体150.5cM处。通过ORF、启动子及基因预测软件分析,Ben可能是编码某种受体蛋白的基因或者催化6-OH-苯达松与葡萄糖缀合、类似淀粉合成酶的基因,而ben可能是Ben的调控因子如启动子、内含子等突变而致。
【Abstract】 The PCR primer sets were designed according to contig 10968 from a draftsequence of the rice genome(Oryza sativa L ssp. indica). The RAPD markers OPG18/972and OPG18/943 cosegragatied with Ben/ben genes were successfully walked to 8138bp and8103bp. OPG18/972 and OPG18/943 locus, and Ben/ben were anchored to chromosome 2at 150cM in silico. Open reading frame, promoter and Ben/ben were predicted. Theresults give a clue that Ben gene maybe code a receptor protein or code a certain starchsynthase--like which metabolizes 6--hydroxybentazon (6--OH--bentazon) to the glucoseconjugate. The change of Ben gene at regulatory elements (such as promoter, intron etc)perhaps produced the ben gene.
