RAPD技术在辣椒品种资源遗传分析中的初步应用

【摘要】 用30个10bp的随机引物对30个国内外辣椒品种进行PCR扩增,筛选出28个多态性引物,共扩增出111条带,平均每引物可扩增条带3.96条,其中多态性条带数90条,平均每个引物产生3.21个多态性条带。将RAPD扩增结果构建Neighbor-Joining树形图,并结合辣椒品种的性状进行分析,结果发现:聚类图上显示的分类结果与辣椒果味或者果形之间总体上并无相关性。但在某些小组内,辣味品种内部或甜椒品种内部的遗传相似性高于这两种类型之间的遗传相似性;灯笼形辣椒和角形(羊角形和牛角形)辣椒品种内的遗传相似性也高于这两类品种之间的遗传相似性。试验结果为辣椒种质资源的鉴定以及优良基因的开发利用提供了一定的参考依据。

【Abstract】 Total 30 DNA samples from Capsicum were analyzed by RAPD andtwenty--eight out of thirty 10--base RAPD primers used gave out polymorphism. 90 bands,81% of total 111 amplified bands were divergent with an averaged 3.21 polymorphic bands per primer.A genetic dendrogram among the materials was constructed by Neighbor-joining method with MEGA software based on the calculated similarity coefficient. Theresult showed that taxonomical results from dendrogram did not correlate with fruit shapeand fruit taste,with some exceptions in some subgroups, where divergence of intratypesin sweet or pungent peppers was less than intertypes. It was the same with the geneticrelationships among horn--shape cultivars or bell(shape peppers. Analysis of the geneticrelationships among the germplasms would provide a theoretical ground for Capsicumbreeding or other related researches.
